This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 62 of the PLOD1 protein (p.Glu62Lys). This variant is present in population databases (rs146092290, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with PLOD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 292279). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

This frequency is not significantly higher than estimated for a pathogenic variant in PLOD1 causing Ehlers-Danlos syndrome, kyphoscoliotic type 1, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.184G>A in individuals affected with Ehlers-Danlos syndrome, kyphoscoliotic type 1 and no experimental evidence demonstrating its impact on protein function have been reported.
