PTEN hamartoma tumor syndrome Pathogenic:1
This sequence change deletes 3 nucleotide and inserts 2 nucleotides in exon 5 of the PTEN mRNA (c.485_487delinsCC), causing a frameshift at codon 162. This creates a premature translational stop signal (p.Asp162Alafs*5) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in PTEN are known to be pathogenic (PMID: 9467011, 21194675). For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.485_487delACAinsCC pathogenic mutation, located in coding exon 5 of the PTEN gene, results from the deletion of 3 nucleotides and insertion of two nucleotides causing a translational frameshift with a predicted alternate stop codon (p.D162Afs*5). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
