Variant summary: CASR c.1622A>G (p.Asn541Ser) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251488 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1622A>G in individuals affected with Autosomal Dominant Hypocalcemia and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 532578). Based on the evidence outlined above, the variant was classified as uncertain significance. -
Nephrolithiasis/nephrocalcinosis Uncertain:1
The p.N541S variant (also known as c.1622A>G), located in coding exon 5 of the CASR gene, results from an A to G substitution at nucleotide position 1622. The asparagine at codon 541 is replaced by serine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
