NM_000388.4:c.2104G>C
Variant summary
Our verdict is Likely pathogenic. The variant received 7 ACMG points: 7P and 0B. PM1PM2PP2PP3_Moderate
The NM_000388.4(CASR):c.2104G>C(p.Val702Leu) variant causes a missense change. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. V702I) has been classified as Uncertain significance.

The p.V702L variant (also known as c.2104G>C), located in coding exon 6 of the CASR gene, results from a G to C substitution at nucleotide position 2104. The valine at codon 702 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
