This variant disrupts the p.Glu412 amino acid residue in EGR2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 17717711, 27013732). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individual(s) with Charcot-Marie-Tooth disease (PMID: 22546699, 30843326). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 409979). This variant is present in population databases (rs749558026, ExAC 0.002%). This sequence change replaces glutamic acid with glycine at codon 412 of the EGR2 protein (p.Glu412Gly).
