The c.530G>A (p.R177H) alteration is located in exon 3 (coding exon 3) of the SERPINC1 gene. This alteration results from a G to A substitution at nucleotide position 530, causing the arginine (R) at amino acid position 177 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. In summary, this is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a SERPINC1-related disease. This sequence change replaces arginine with histidine at codon 177 of the SERPINC1 protein (p.Arg177His). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and histidine.
