Variant summary: CFTR c.1366G>T (p.Val456Phe) results in a non-conservative amino acid change located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 244512 control chromosomes. c.1366G>T has been reported in the literature in individuals affected with Cystic Fibrosis (examples: Dork_1994a, Geurts_2020, Dork_1994b, Terzic_2019). These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.1367T>C, p.Val456Ala), supporting the critical relevance of codon 456 to CFTR protein function. The following publications have been ascertained in the context of this evaluation (PMID: 23420618, 7525450, 7505767, 32084388, 12732620, 31523618).
