This missense change has been observed in individual(s) with clinical features of CFTR-related conditions (PMID: 32357917). ClinVar contains an entry for this variant (Variation ID: 495898). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant disrupts the p.Pro499 amino acid residue in CFTR. Other variant(s) that disrupt this residue have been observed in individuals with CFTR-related conditions (PMID: 9630075; Invitae), which suggests that this may be a clinically significant amino acid residue. The p.P499S variant (also known as c.1495C>T), located in coding exon 11 of the CFTR gene, results from a C to T substitution at nucleotide position 1495. The proline at codon 499 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
