Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant allele was found at a frequency of 3.2e-05 in 251446 control chromosomes (gnomAD). c.653T>A has been reported in the literature in multiple homozygous and heterozygous individuals affected with Cystic Fibrosis (e.g.McCormick_2002, Mei-Zahav_2005, Sharma_2009, Zampoli_2021). These data indicate that the variant is very likely to be associated with disease. Three submitters have provided assessments for this variant to ClinVar after 2014 and all classified the variant as pathogenic.
