NM_000530.8:c.380_383dupGTGA
Variant summary
Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_000530.8(MPZ):c.380_383dupGTGA(p.Asp128GlufsTer2) variant causes a frameshift, stop gained change. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. D128D) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

Submissions by phenotype
Charcot-Marie-Tooth disease, type I Pathogenic:1
This sequence change inserts 4 nucleotides in exon 3 of the MPZ mRNA (c.380_383dupGTGA), causing a frameshift at codon 128. This creates a premature translational stop signal (p.Asp128Glufs*2) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in MPZ are known to be pathogenic (PMID: 14711881). For these reasons, this variant has been classified as Pathogenic.
