The encoded protein is involved in the urea cycle which functions to detoxify ammonia into urea for excretion. This variant has been observed in individual(s) with ornithine transcarbamylase deficiency (Invitae). In at least one individual the variant was observed to be de novo. This sequence change replaces proline with serine at codon 169 of the OTC protein (p.Pro169Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OTC protein function. This variant disrupts the p.Pro169 amino acid residue in OTC. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 11793468, 11117428, 26059767). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -
