Variant summary: PMS2 c.113C>A (p.Ala38Glu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 245784 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.113C>A in individuals affected with Lynch Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1025441). Based on the evidence outlined above, the variant was classified as uncertain significance.

This sequence change replaces alanine with glutamic acid at codon 38 of the PMS2 protein (p.Ala38Glu). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and glutamic acid.

The p.A38E variant (also known as c.113C>A), located in coding exon 2 of the PMS2 gene, results from a C to A substitution at nucleotide position 113. The alanine at codon 38 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
