Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251252 control chromosomes (gnomAD). c.842_849dupTCCCCGCA has been reported in the literature in 1 homozygote and 5 compound heterozygotes affected with Niemann-Pick Disease Type A (Zhang_2013). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
