The p.D439E variant (also known as c.1317C>A), located in coding exon 12 of the TSC2 gene, results from a C to A substitution at nucleotide position 1317. The aspartic acid at codon 439 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, glutamic acid is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
