NM_000548.5:c.2974C>T
Variant summary
Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_000548.5(TSC2):c.2974C>T(p.Gln992*) variant causes a stop gained change. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. Q992Q) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

The p.Q992* pathogenic mutation (also known as c.2974C>T), located in coding exon 26 of the TSC2 gene, results from a C to T substitution at nucleotide position 2974. This changes the amino acid from a glutamine to a stop codon within coding exon 26. This mutation has been detected in the literature in two individuals with clinical diagnoses or high suspiscions of having tuberous sclerosis complex (TSC) (Beauchamp RL et al. Hum. Mutat., 1998;12:408-16; Rosset C et al. PLoS ONE, 2017 Oct;12:e0185713). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
