The c.109C>G (p.P37A) alteration is located in exon 1 (coding exon 1) of the FASLG gene. This alteration results from a C to G substitution at nucleotide position 109, causing the proline (P) at amino acid position 37 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Autoimmune lymphoproliferative syndrome type 1 Uncertain:1
This sequence change replaces proline with alanine at codon 37 of the FASLG protein (p.Pro37Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. This variant is present in population databases (rs145731782, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with FASLG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
