Glycogen storage disease type III Pathogenic:1
This sequence change affects a donor splice site in intron 13 of the AGL gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with glycogen storage disease type III (PMID: 26984562, 29614965). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. Studies have shown that disruption of this splice site results in skipping of exon 13, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 8702417). For these reasons, this variant has been classified as Pathogenic. -
