The c.11891C>T (p.S3964L) alteration is located in exon 89 (coding exon 89) of the RYR3 gene. This alteration results from a C to T substitution at nucleotide position 11891, causing the serine (S) at amino acid position 3964 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine with leucine at codon 3964 of the RYR3 protein (p.Ser3964Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs750201337, ExAC 0.01%). This variant has not been reported in the literature in individuals with RYR3-related disease.
