Submissions by phenotype
not specified Uncertain:1
The c.160T>G (p.S54A) alteration is located in exon 2 (coding exon 2) of the RYR3 gene. This alteration results from a T to G substitution at nucleotide position 160, causing the serine (S) at amino acid position 54 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Epileptic encephalopathy Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with RYR3-related disease. This variant is present in population databases (rs771903948, ExAC 0.002%). This sequence change replaces serine with alanine at codon 54 of the RYR3 protein (p.Ser54Ala). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and alanine. -
