This variant was observed as part of a predisposition screen in an ostensibly healthy population. A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). No publications were found based on this search. Allele frequency data from public databases did not allow this variant to be ruled in or out of causing disease. Therefore, this variant is classified as a variant of unknown significance. -
This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1035 of the MLH3 protein (p.Glu1035Lys). This variant is present in population databases (rs143865811, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with MLH3-related conditions. ClinVar contains an entry for this variant (Variation ID: 566416).
