The p.I1676V variant (also known as c.5026A>G), located in coding exon 26 of the SCN2A gene, results from an A to G substitution at nucleotide position 5026. The isoleucine at codon 1676 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -

This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1676 of the SCN2A protein (p.Ile1676Val). This variant is present in population databases (rs755159786, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with SCN2A-related conditions. ClinVar contains an entry for this variant (Variation ID: 574402).
