The NF1 c.2251+2T>C variant, also published as IVS13+2T>C, is reported in the literature in several individuals affected with neurofibromatosis type 1 (Pros 2008, Serra 2001). This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. This variant disrupts the canonical splice donor site of intron 18 (also published as intron 13), which is likely to negatively impact gene function. Indeed, cDNA analysis in patient fibroblasts suggests this variants results in exon skipping and a frameshift (Pros 2008). Based on available information, this variant is considered to be pathogenic.
