The p.Q950* pathogenic mutation (also known as c.2848C>T), located in coding exon 21 of the NF1 gene, results from a C to T substitution at nucleotide position 2848. This changes the amino acid from a glutamine to a stop codon within coding exon 21. This mutation has been reported in subjects with features of neurofibromatosis type 1 (Zhang J et al. Sci Rep, 2015 Jun;5:11291; Stella A et al. Genes (Basel), 2018 Apr;9:). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
