The NF1 c.5902C>T variant is predicted to result in premature protein termination (p.Arg1968*). This variant, referred to as c.5839C>T (p.Arg1947*) in an alternate transcript (NM_000267.3), has been reported in multiple individuals with neurofibromatosis type 1 (see for example - Fashold et al. 2000. To our knowledge, this variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. This variant is interpreted as pathogenic in Clinvar (https://www.ncbi.nlm.nih.gov/clinvar/variation/343/). Nonsense variants in NF1 are expected to be pathogenic.
