This variant has been identified in 1/66628 European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitu te.org; dbSNP rs200189282). Computational prediction tools and conservation ana lysis do not provide strong support for or against an impact to the protein. In summary, the clinical significance of the p.Arg1383Trp variant is uncertain. -

This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 1383 of the LAMA4 protein (p.Arg1383Trp). This variant is present in population databases (rs200189282, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with LAMA4-related conditions. ClinVar contains an entry for this variant (Variation ID: 44382). The p.R1383W variant (also known as c.4147C>T), located in coding exon 30 of the LAMA4 gene, results from a C to T substitution at nucleotide position 4147. The arginine at codon 1383 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear. -
