NM_001134363.3:c.1013T>C
Variant summary
Our verdict is Benign. The variant received -7 ACMG points: 0P and 7B. BP4_ModerateBP6BS2
The NM_001134363.3(RBM20):c.1013T>C(p.Met338Thr) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.0000165 in 1,398,132 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. 14/20 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. M338R) has been classified as Uncertain significance.

not specified Uncertain:1
The p.Met338Thr variant in RBM20 has not been previously reported in individuals with cardiomyopathy and was absent from large population studies. Computational prediction tools and conservation analysis suggest that the p.Met338Thr variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, the clinical significance of the p.Met338Th r variant is uncertain. -
Cardiovascular phenotype Uncertain:1
The p.M338T variant (also known as c.1013T>C), located in coding exon 2 of the RBM20 gene, results from a T to C substitution at nucleotide position 1013. The methionine at codon 338 is replaced by threonine, an amino acid with similar properties. This variant co-occurred with a TTN A-band truncating variant and variants in other cardiac-related genes in two affected individuals from a family with dilated cardiomyopathy (Cowan JR et al. Circ Genom Precis Med, 2018 Jul;11:e002038). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
