The p.R163Q variant (also known as c.488G>A), located in coding exon 2 of the RBM20 gene, results from a G to A substitution at nucleotide position 488. The arginine at codon 163 is replaced by glutamine, an amino acid with highly similar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
