This variant is present in population databases (rs142450703, gnomAD 0.1%). This missense change has been observed in individuals with clinical features of EYS-related conditions (PMID: 20333770, 31456290, 35836572; internal data). ClinVar contains an entry for this variant (Variation ID: 551873). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt EYS protein function with a negative predictive value of 80%. Variant summary: EYS c.1852G>A (p.Gly618Ser) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 0.00011 in 157080 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in EYS causing Retinitis Pigmentosa (0.00011 vs 0.0034), allowing no conclusion about variant significance. c.1852G>A has been observed in individuals affected with Retinitis Pigmentosa (e.g., Audo_2010, Gonzalez-delPozo_2011, Maltese_2022, Placidi_2023, Sharon_2020, internal data). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
