This sequence change replaces alanine with valine at codon 1724 of the SCN1A protein (p.Ala1724Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with SCN1A-related conditions (PMID: 31302675, 31273778). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 530420). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant disrupts the p.Ala1724 amino acid residue in SCN1A. Other variant(s) that disrupt this residue have been observed in individuals with SCN1A-related conditions (PMID: 28012175, 30185235), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
