The p.A3748G variant (also known as c.11243C>G), located in coding exon 44 of the TTN gene, results from a C to G substitution at nucleotide position 11243. The alanine at codon 3748 is replaced by glycine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Variant summary: TTN c.10361-2541C>G is located at a position not widely known to affect splicing. This variant corresponds to c.12332C>G (p.Ala4111Gly) in NM_001267550. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 5e-05 in 1613892 control chromosomes, predominantly at a frequency of 0.00093 within the African or African-American subpopulation in the gnomAD database. The observed variant frequency within African or African-American control individuals in the gnomAD database is approximately 2.4-fold of the estimated maximal expected allele frequency for a pathogenic variant in TTN causing dilated cardiomyopathy phenotype (0.00039). To our knowledge, no occurrence of c.10361-2541C>G in individuals affected with dilated cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
