Variant summary: TTN c.30124G>A (p.Glu10042Lys) results in a conservative amino acid change in the encoded protein sequence. Two of three in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00025 in 220756 control chromosomes, predominantly at a frequency of 0.00048 within the Non-Finnish European subpopulation in the gnomAD database. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database is approximately 1.2 fold of the estimated maximal expected allele frequency for a pathogenic variant in TTN causing Autosomal Recessive Titinopathy phenotype (0.00039), suggesting the variant may be benign. c.30124G>A has been reported in the literature in at least one individual affected with Dilated Cardiomyopathy in the presence of multiple variants, without evidence of causality (e.g. Forleo_2017), or in an individual affected with Bannayan-Riley-Ruvalcaba syndrome without evidence for causality (e.g. Yehia_2017). These report(s) do not provide unequivocal conclusions about association of the variant with Autosomal Recessive Titinopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 28750076, 29263846). ClinVar contains an entry for this variant (Variation ID: 46895). Based on the evidence outlined above, the variant was classified as likely benign. -
