In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function; Has not been previously published as pathogenic or benign to our knowledge -
MEGF8: BP4 -
MEGF8-related Carpenter syndrome Uncertain:1
This sequence change replaces proline with leucine at codon 2100 of the MEGF8 protein (p.Pro2100Leu). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and leucine. This variant is present in population databases (rs372350131, ExAC 0.1%). This variant has not been reported in the literature in individuals affected with MEGF8-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Inborn genetic diseases Uncertain:1
The c.6299C>T (p.P2100L) alteration is located in exon 36 (coding exon 36) of the MEGF8 gene. This alteration results from a C to T substitution at nucleotide position 6299, causing the proline (P) at amino acid position 2100 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
MEGF8-related disorder Uncertain:1
The MEGF8 c.6299C>T variant is predicted to result in the amino acid substitution p.Pro2100Leu. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.13% of alleles in individuals of South Asian descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
