ClinVar contains an entry for this variant (Variation ID: 506138). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt FHL2 protein function with a negative predictive value of 80%. A different nucleotide substitution affecting the same amino acid (p.Arg131Cys) has been reported in an individual with dilated cardiomyopathy (Table S1B, Walsh et al. 2017. PubMed ID: 27532257). At this time, the clinical significance of the c.392G>A (p.Arg131His) variant is uncertain due to the absence of conclusive functional and genetic evidence. -
