Inborn genetic diseases Uncertain:1
The c.325G>C (p.V109L) alteration is located in exon 2 (coding exon 1) of the NRXN1 gene. This alteration results from a G to C substitution at nucleotide position 325, causing the valine (V) at amino acid position 109 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Pitt-Hopkins-like syndrome 2 Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The leucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. ClinVar contains an entry for this variant (Variation ID: 539875). This variant has not been reported in the literature in individuals affected with NRXN1-related conditions. This variant is present in population databases (rs779979011, gnomAD 0.006%). This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 109 of the NRXN1 protein (p.Val109Leu). -
