This sequence change creates a premature translational stop signal (p.Tyr351*) in the MEN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MEN1 are known to be pathogenic (PMID: 12112656, 17853334). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of MEN1-related conditions (PMID: 16563611). ClinVar contains an entry for this variant (Variation ID: 428089). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.

The p.Y351* pathogenic mutation (also known as c.1053C>G), located in coding exon 7 of the MEN1 gene, results from a C to G substitution at nucleotide position 1053. This changes the amino acid from a tyrosine to a stop codon within coding exon 7. This mutation has been previously reported in a Danish family affected with multiple endocrine neoplasia type 1 (MEN1) (Jäger AC, Mol. Cell. Endocrinol. 2006 Apr; 249(1-2):123-32). In addition to the clinical data presented in the literature, since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294).
