This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 368 of the CCDC78 protein (p.Gly368Glu). This variant is present in population databases (rs145971446, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with CCDC78-related conditions. ClinVar contains an entry for this variant (Variation ID: 473249). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CCDC78 protein function with a negative predictive value of 80%.
