This gene is expressed in the inner ear of vertebrates with the highest level of expression seen at the embryonic stage and lowest in adult. Knockdown studies in zebrafish suggest that this gene is essential for normal inner ear function. Mutations in this gene are associated with autosomal recessive deafness. This sequence change replaces glutamine, which is neutral and polar, with lysine, which is basic and polar, at codon 1117 of the OTOGL protein (p.Gln1117Lys). This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with OTOGL-related conditions. ClinVar contains an entry for this variant (Variation ID: 229117). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. The p.Gln1117Lys variant in OTOGL has not been previously reported in individual s with hearing loss or in large population studies. Glutamine (Gln) at position 1117 is not conserved in fish, of which 13 species carry a lysine (Lys) at this position, raising the possibility that this change may be tolerated. Additional computational prediction tools do not provide strong support for or against an i mpact to the protein. In summary, the clinical significance of the p.Gln1117Lys variant is uncertain. -
