NM_001387283.1:c.672G>T
Variant summary
Our verdict is Uncertain significance. The variant received 1 ACMG points: 2P and 1B. PM2BP6
The NM_001387283.1(SMARCA4):c.672G>T(p.Ser224Ser) variant causes a synonymous change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Synonymous variant affecting the same amino acid position (i.e. S224S) has been classified as Likely benign.

Hereditary cancer-predisposing syndrome Uncertain:1
The c.672G>T variant (also known as p.S224S), located in coding exon 3 of the SMARCA4 gene, results from a G to T substitution at nucleotide position 672. This nucleotide substitution does not change the serine at codon 224. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. Based on the available evidence, the clinical significance of this variant remains unclear.
