The homozygous p.Arg228Ter variant in B4GALNT1 was identified by our study in one individual with Spastic Paraplegia. This variant has been reported as homozygous in the literature in two affected family members (Boukhris et al. 2013; PMID: 23746551). This variant has been identified in <0.01% (1/33582) of Latino chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs398122382). Although this variant has been seen in the general population, its frequency is low enough to be consistent with a recessive carrier frequency. Loss of function of the B4GALNT1 gene is an established disease mechanism in autosomal recessive Spastic Paraplegia, and this is a loss of function variant. In summary, this variant is pathogenic. -
