GARS1 (HGNC:4162): (glycyl-tRNA synthetase 1) This gene encodes glycyl-tRNA synthetase, one of the aminoacyl-tRNA synthetases that charge tRNAs with their cognate amino acids. The encoded enzyme is an (alpha)2 dimer which belongs to the class II family of tRNA synthetases. It has been shown to be a target of autoantibodies in the human autoimmune diseases, polymyositis or dermatomyositis. The GARS1 c.1234C>T variant is predicted to result in the amino acid substitution p.Arg412Cys. This variant was reported in a compound heterozygous individual with mitochondrial respiratory chain dysfunction who presented with left ventricular wall hypertrophy, lactic acidosis, and exercise intolerance (Nafisinia et al 2017. PubMed ID: 28594869). This variant is reported in 0.0058% of alleles in individuals of Latino descent in gnomAD. While we suspect this variant could be pathogenic, at this time the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
