This sequence change replaces cysteine with arginine at codon 442 of the HSPD1 protein (p.Cys442Arg). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with HSPD1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). -

The c.1324T>C (p.C442R) alteration is located in exon 10 (coding exon 9) of the HSPD1 gene. This alteration results from a T to C substitution at nucleotide position 1324, causing the cysteine (C) at amino acid position 442 to be replaced by an arginine (R). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
