This variant is present in population databases (rs587781530, gnomAD 0.003%). This missense change has been observed in individual(s) with a personal and/or family history of breast cancer, ovarian cancer, and/or bladder cancer (PMID: 30613976, 31159747, 35534704, 36346689). ClinVar contains an entry for this variant (Variation ID: 141146). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The p.S40L variant (also known as c.119C>T), located in coding exon 2 of the NBN gene, results from a C to T substitution at nucleotide position 119. The serine at codon 40 is replaced by leucine, an amino acid with dissimilar properties. This alteration has been reported as variant of uncertain significance in 1/1197 individuals from Greece, Romania, and Turkey undergoing evaluation for inherited cancer predisposition (Tsaousis GN et al. BMC Cancer, 2019 Jun;19:535). This alteration was also identified in individuals diagnosed with breast cancer (Hauke J et al. Cancer Med, 2018 04;7:1349-1358; Rizzolo P et al. Int J Cancer, 2019 07;145:390-400). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
