Mutations in this gene have been associated with hereditary sensory and autonomic neuropathy, type 5 (HSAN5), and dysregulation of this gene's expression is associated with allergic rhinitis. This sequence change replaces proline with histidine at codon 112 of the NGF protein (p.Pro112His). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and histidine.
