NTRK1 (HGNC:8031): (neurotrophic receptor tyrosine kinase 1) This gene encodes a member of the neurotrophic tyrosine kinase receptor (NTKR) family. This kinase is a membrane-bound receptor that, upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. The presence of this kinase leads to cell differentiation and may play a role in specifying sensory neuron subtypes. Mutations in this gene have been associated with congenital insensitivity to pain, anhidrosis, self-mutilating behavior, cognitive disability and cancer. The NTRK1 c.295G>A; p.Val99Met variant (rs201509045, ClinVar Variation ID: 456616) is reported in the literature in an individuals affected with prostate cancer, though no other evidence of causality was presented (Liang 2022). This variant is found in the East Asian population with an allele frequency of 0.16% (32/19,942 alleles) in the Genome Aggregation Database (v2.1.1). Computational analyses are uncertain whether this variant is neutral or deleterious (REVEL: 0.335). Due to limited information, the clinical significance of this variant is uncertain at this time.
