This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 357 of the POLD1 protein (p.Leu357Arg). This variant is present in population databases (rs143340270, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 408004). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt POLD1 protein function with a positive predictive value of 80%. The p.L357R variant (also known as c.1070T>G), located in coding exon 8 of the POLD1 gene, results from a T to G substitution at nucleotide position 1070. The leucine at codon 357 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
