This sequence change replaces methionine, which is neutral and non-polar, with lysine, which is basic and polar, at codon 937 of the POLD1 protein (p.Met937Lys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 937513). The p.M937K variant (also known as c.2810T>A), located in coding exon 21 of the POLD1 gene, results from a T to A substitution at nucleotide position 2810. The methionine at codon 937 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
