The POLD1 c.934G>A (p.Val312Met) variant has been reported in the published literature as one of several germline variants in an individual with colorectal cancer (PMIDs: 31769227 (2020)) and in an individual with obsessive-compulsive disorder (PMID: 34183866 (2021)). The frequency of this variant in the general population, 0.000056 (7/125888 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -

This variant is present in population databases (rs371612922, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 246387). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLD1 protein function with a negative predictive value of 80%.
