The p.Ser171Arg variant in POU4F3 has not been previously reported in individual s with hearing loss, but has been identified in 13/65634 European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP r s148985828). Although this variant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. Computational predic tion tools and conservation analyses do not provide strong support for or agains t an impact to the protein. In summary, the clinical significance of this varian t is uncertain.

This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 171 of the POU4F3 protein (p.Ser171Arg).
