NM_002878.4:c.324C>G
Variant summary
Our verdict is Likely benign. The variant received -2 ACMG points: 2P and 4B. PM2BP4_ModerateBP6BP7
The NM_002878.4(RAD51D):c.324C>G(p.Gly108Gly) variant causes a synonymous change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Synonymous variant affecting the same amino acid position (i.e. G108G) has been classified as Likely benign.

Hereditary cancer-predisposing syndrome Uncertain:1
The c.324C>G variant (also known as p.G108G), located in coding exon 4 of the RAD51D gene, results from a C to G substitution at nucleotide position 324. This nucleotide substitution does not change the amino acid at codon 108. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive and direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
Breast-ovarian cancer, familial, susceptibility to, 4 Benign:1
This variant is considered benign. This variant is a silent/synonymous amino acid change and it is not expected to impact splicing. -
