This gene encodes a protein found in striated and cardiac muscle that binds to the titin Z1-Z2 domains and is a substrate of titin kinase, interactions thought to be critical to sarcomere assembly. This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 153 of the TCAP protein (p.Arg153Pro). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TCAP-related conditions.
